RESUMO

Different methods of purification and drying of sericin were evaluated to obtain a material free of contaminating residues through the development of safer and sustainable environmental technology. Samples purified using different proportions of alcohol solvents and by freezing/thawing were compared, while drying was performed by lyophilisation and atomisation in a spray. No significant statistical differences were observed in the yield of the purification process compared with the evaluated methods. Thus, the freezing/thawing method has economic and environmental advantages over other methods. Drying by spraying and lyophilisation produced particles exhibiting different properties in terms of structure: ß-sheets, random sheets, and ß-turns. In general, these particles are amorphous. The particles obtained by spray drying were more luminous, soluble, and spherical. Thus, purification by freezing/thawing and drying by spray can be considered appropriate methodologies for obtaining contaminant-free sericin through sustainable environmental technology.

RESUMO

Natural products can be used as complements or as alternatives to synthetic drugs. Eugenia uniflora and Tropaeolum majus are natives of Brazil and have antimicrobial, anti-inflammatory, and antioxidant activities. This study aimed to develop a film-forming system (FFS) loaded with plant extracts with the potential for treating microbial infections. E. uniflora and T. majus leaf extracts were prepared and characterized, and the individual and combined antioxidant and antimicrobial activities were evaluated. The FFS was developed with different concentrations of polyvinylpyrrolidone (PVP) and polyvinyl alcohol (PVA) and analyzed for physicochemical characteristics. The combination of extracts showed a superior antioxidant effect compared to the individual extracts, justifying the use of the blend. FFS prepared with 4.5% PVA, 4.5% PVP, 7.81% E. uniflora extract, and 3.90% T. majus extract was adhesive, lacked scale formation, presented good malleability, and had a suitable pH for topical application. In addition, the viscosity at rest was satisfactory for maintaining stability; water solubility was adequate; skin permeation was low; and the antimicrobial effect was superior to that of the individual extracts. Therefore, the developed FFS is promising for the differentiated treatment of skin lesions through topical application.

RESUMO

Plinia cauliflora (Mart.) Kausel, popularly known as jabuticaba, possesses bioactive compounds such as flavonoids, tannins, and phenolic acids, known for their antioxidant, antibacterial, wound healing, and cardioprotective effects. Therefore, this study aimed to standardize the P. cauliflora fruit peel extraction method, maximize phenolic constituents, and evaluate their antioxidative and antimicrobial effects. Various extraction methods, including vortex extraction with and without precipitation at 25, 40, and 80 °C, and infusion extraction with and without precipitation, were performed using a completely randomized design. Extraction without precipitation (E - P) showed the highest yield (57.9%). However, the precipitated extraction (E + P) method displayed a yield of 45.9%, higher levels of phenolic derivatives, and enhanced antioxidant capacity. Major compounds, such as D-psicose, D-glucose, and citric acid, were identified through gas chromatography-mass spectrometry (GC-MS) analysis. Ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) analysis identified citric acid, hexose, flavonoids, tannins, and quercetin as the major compounds in the extracts. Furthermore, the extracts exhibited inhibitory effects against Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli bacteria. In conclusion, the E + P method efficiently obtained extracts with high content of bioactive compounds showing antioxidant and antimicrobial capacities with potential application as a dietary supplement.

RESUMO

Chagas disease is a neglected tropical disease with only two drugs available for treatment and the plant Cecropia pachystachya has several compounds with antimicrobial and anti-inflammatory activities. This study aimed to evaluate a supercritical extract from C. pachystachya leaves in vitro and in vivo against Trypanosoma cruzi. A supercritical CO2 extraction was used to obtain the extract (CPE). Cytotoxicity and immunostimulation ability were evaluated in macrophages, and the in vitro trypanocidal activity was evaluated against epimastigotes and trypomastigotes forms. In vivo tests were done by infecting BALB/c mice with blood trypomastigotes forms and treating animals orally with CPE for 10 days. The parasitemia, survival rate, weight, cytokines and nitric oxide dosage were evaluated. CPE demonstrated an effect on the epi and trypomastigotes forms of the parasite (IC50 17.90 ± 1.2 µg/mL; LC50 26.73 ± 1.2 µg/mL) and no changes in macrophages viability, resulting in a selectivity index similar to the reference drug. CPE-treated animals had a worsening compared to non-treated, demonstrated by higher parasitemia and lower survival rate. This result was attributed to the anti-inflammatory effect of CPE, demonstrated by the higher IL-10 and IL-4 values observed in the treated mice compared to the control ones. CPE demonstrated a trypanocidal effect in vitro and a worsening in the in vivo infection due to its anti-inflammatory activity.

Beauty is directly linked to the appearance of hair, in all times and all cultures. Hair throughout life is subject to various types of changes, whether natural or artificial, driven by a desire to change, often because it represents a new phase, as it is also a symbol of self-care, vanity, success, they have significant importance in people's appearance and self-esteem and, therefore, hair loss can affect the individual's quality of life. In this sense, the objective of this study was to evaluate minimally invasive and non- surgical aesthetic procedures with an emphasis on hair loss. To this end, an integrative bibliographic review was carried out, of a descriptive nature, in the main health databases: SciELO, Google Scholar, PubMed and medical books, in Portuguese, English and Spanish, considering the period from 2007 to 2022. According to the latest census by the Brazilian Society of Dermatology, the complaint of alopecia is among the ten most frequent complaints in dermatological offices. The most important hair diseases are divided into two groups, scarring when hair loss is accompanied by scars resulting in permanent baldness, and non-scarring where baldness is not permanent leading to gradual thinning and thinning of the hair. Non-scarring alopecia is the most frequent and the most common among them are androgenetic alopecia and telogen effluvium. Many patients seek treatment to improve their appearance and self-esteem, which may be related to anxiety and depression. These treatments must be individualized, where the individual characteristics of each patient must be evaluated. Aesthetic hair treatments can contribute significantly to reducing low self-esteem, contributing to the recovery of self-confidence through hair growth provided by aesthetics. Therefore, it is concluded that the results collected in the literature review were satisfactory, in a way that shows that medicines, supplements and aesthetic treatments in hair loss have been evolving more and more. However, more studies on the subject are needed.

Through the edition of the Operational Norm for Health Care carried out in 2001 and the publication of the Pact for Health carried out in 2006, the process of access to health gained greater emphasis on the issue of innovations and improvements in the health system. Pharmaceutical assistance emerged as a fundamental part of health services and programs. In this sense, the objective of the present study was to expose the main public policies on the subject of essential medicines. Therefore, an integrative literature review was carried out, using the Ministry of Health, the National Health Surveillance Agency's Resolutions, scientific articles and the publications of the National Medicines List (RENAME) as a database. In 1998, through ordinance nº 3.916, of October 30, 1998, the public policy terminology is presented to the population and, in response to the guidelines resulting from this policy, in this article we will emphasize RENAME, created through ordinance nº 3.047 , of November 28, 2019, which meets the basic and fundamental principles of the Unified Health System: universality, equity and integrality, to meet the treatments of the various diseases and conditions that affect the Brazilian population. Therefore, it is concluded that it is not just about public policies, but about maintaining the right of the citizen established by the Constitution of the Federative Republic of Brazil, so that the patient can acquire the appropriate medication and in the necessary quantity, allowing professionals to achieve more use in the management of the pharmaceutical care cycle.

Thus, the objective of this work was to emphasize the importance of the cultivation and rational use of herbal medicines and medicinal plants. To this end, articles in Portuguese and English inserted in the database: Scielo, Google academic, in addition to electronic journals, books with scientific reference base and database of the Ministry of Health between the years 2018 to 2022 were included. herbal medicines can cause adverse effects, interfere with the effect of medicines used concomitantly, in addition to the possibility of causing poisoning due to the presence of contaminants in low-quality products. For the production of bioactives, at qualitatively and quantitatively appropriate levels, the cultivation of medicinal plants must be carefully carried out, through techniques cultivated for soil preservation and planting are essential, such as the use of green manures and vegetation cover, which in addition to protect the soil from solar radiation, prevent excessive water evaporation and improve the physical, chemical and biological characteristics of the sun. These criteria influence the quality of the herbal medicine, therefore, in order to guarantee therapeutic efficacy, as the plants must be correctly cultivated, collected, identified and conserved. Therefore, it is concluded that the use of medicinal plants for treatments has been increasingly indicated by health professionals, emphasizing the importance of safe and rational use, with an alert to its consequences.

RESUMO

Medicinal plants have great prominence in research into the development of new medicines. Eugenia uniflora L. (Myrtaceae) is an edible and medicinal plant with economic value in the northeast region of Brazil. Several preparations from E. uniflora leaves and its fruits are employed as a source of nutrients and bioactive compounds. In this study we evaluated the preclinical toxicology of crude extract and vaginal gel obtained from the leaves of E. uniflora (5%, 10%, and 15%) aiming to provide safety for its use in the treatment of vulvovaginitis. Both formulations were applied to the vaginal cavity for 14 days. Detailed observations of the vaginal region, including pruritus, swelling, irritation, burning, pain, and vaginal secretion, as well as the estrous cycle were evaluated. On the fifth day, blood samples were obtained from the supraorbital plexus for biochemical and hematological analyses. The animals were subsequently euthanized. All animals underwent necropsy and macroscopic examination of the vaginal mucosa and reproductive system. A histological examination was also performed. No clinically significant changes were detected during the entire experimental period. All biochemical, hematological, or histopathological parameters were within the normal range for the species. The data obtained allow us to suggest that the E. uniflora vaginal formulations are safe in this experimental model.

Studies have already demonstrated the antioxidant, anti-inflammatory and antidiabetic potential of this extract, where phenols, lignans, tannins and flavonoids were identified. The objective of this work was to optimize the extractive method of A. lappa roots. Extracts were prepared by different methods: Ultrasound, Soxhlet, maceration and vortical extraction. The optimization was performed by vortical extraction following a 23 full factorial design, using as factors: alcohol content, drug concentration and extraction time. The extracts were evaluated for dry residue, phenols and flavonoids contents, and antioxidant activity. Regarding the dry residue, and the phenols and flavonoids contents, the ultrasound and vortical extraction methods showed better extractive power. Due to the lower operating time and cost, the optimization was performed by vortical extraction, and the optimized extract was obtained using 60% alcohol, in a 1:10 drug solvent ratio and extraction time of 15 minutes. These assessments guide the future tests of transposition of the method to an industrial scale, reducing manpower, time and costs, aiming to obtain more efficient phytotherapic products, with affordable value for the population.

Honey is a natural food product, used by the population since ancient Egypt, as a food source and in popular medicine, due to its antioxidant, healing, expectorant, sedative, analgesic and anti-inflammatory properties, as well as antibacterial activity. The increase in honey consumption by the population, associated with the practice of beekeeping, has been shown to be a promising and profitable activity for producers and for the Brazilian economy. However, due to the increase in consumption demand, honey may be the target of the addition of adulterating substances, such as commercial sugar, causing a decrease in product quality. The objective of this work was to perform an analysis of the physicochemical properties of samples of honey marketed in open fairs of the municipality of Assis Chateaubriand-PR. The parameters evaluated were moisture, dye presence, density determination, Jagerschmidt reactions, Lugol, Fiehe, total acidity, ash content and analysis. The samples were analyzed commercially at free trade fairs in the municipality of Assis Chateaubriand. microscopic and organoleptic. For the execution of the experiments, the methodology adopted was based on the physical-chemical methods for Food Analysis, of the Adolf Lutz Institute, being the same, carried out in triplicate. It can be concluded that of the five samples analyzed, two were positive for sugar addition. The presented results obtained variations among the samples, since the composition of the honey differs according to the floral species and region from which it comes from, besides the conditions of storage and handling of the same.

Abstract Pterodon pubescens (Benth.) Benth., Fabaceae, fruits have been investigated for their anti-inflammatory and antinociceptive activities, and have demonstrated effectiveness in inflammatory conditions. A physiochemical and microbiological stability study was conducted to investigate two nanoemulsion-based delivery systems of two different hydrophilic surfactants (polyethylene glycol-40H castor oil or polyethylene glycol-40 castor oil). The nanoemulsions, containing P. pubescens oil, lecithin, hydrophilic surfactant and water, were analyzed for droplet size distribution, polydispersity index, pH, consistency index, stability against centrifugal force, and active content/vouacapan derivatives. The physicochemical characteristics were followed for 365 days. The nanoemulsion system was evaluated for anti-inflammatory activity by using with a peritonitis model, immediately after preparation and after 365 days of storage at 25 °C. The stability study demonstrated that proper storage (25 °C) preserved the characteristics of the nanoemulsion containing 7.5% polyethylene glycol-40H castor oil, 5% lecithin, and 5% P. pubescens oil. Further, it ensured a shelf life of 365 days as a phytotherapeutic formulation. In the peritonitis assay induced by carrageenan, nanoemulsion prepared with polyethylene glycol-40H castor oil (125 mg/kg) reduced leukocyte migration, even after 365 days of storage (25 °C), highlighting its potential for the treatment of inflammatory diseases. However, further studies are needed to confirm its clinical effectiveness.

RESUMO

Currently, leishmaniasis is difficult to manage owing to the limited choice and high toxicity of available drugs, and emergence of drug-resistant protozoa. Medicinal plants, which produce various bioactive molecules, can help counter this global shortage. In this study, we prepared Pterodon pubescens fruit extracts, which show antileishmanial activity, and developed a nanoemulsion of the optimized extract to improve its performance. The extracts were prepared using conventional methods and a supercritical fluid method and were tested for activity against Leishmania amazonensis promastigotes and amastigotes. The two most effective extracts were formulated as nanoemulsions. Although both extracts showed cytotoxicity, the supercritical extracts were more effective against L. amazonensis promastigotes and amastigotes than conventional extracts were. This was attributed to the high content of the geranylgeraniol derivative in the supercritical extracts. The nanoemulsions showed a better selectivity index and significantly improved activity against parasites (IC50: 2.7 µg/mL for nanoemulsion of hexane extract; IC50: 1.9 µg/mL for nanoemulsion of supercritical extract) compared to the Miltefosine standard (0.7 µg/mL). This could be due to the smaller droplets of the supercritical extracts, allowing better penetration. In conclusion, the extracts showed promise in in vitro tests, and could be used as a leishmaniasis treatment.

RESUMO

Trees of the genus Pterodon, commonly known as "sucupira-branca" or "faveira," are native to central Brazil. The Pterodon fruits are traditionally used in ethnomedicine as an infusion, in small doses, and at regular time intervals as an antirheumatic, anti-inflammatory, tonic, and depurative agent. The various compounds present in the Pterodon class are, generally, water-insoluble and derived from the fusion of high-molecular weight pentacarbonate units. Scientific research has shown that the major compounds isolated from Pterodon species are linear and/or tetracyclic diterpenes with vouacapane skeletons that partly underlie the pharmacological activities of the fruit-derived oil. Material from Pterodon species has several biological properties, such as analgesic, anti-inflammatory, and anticancer effects. Therefore, recent studies have sought to microencapsulate these extracts to protect them from potential chemical degradation and improve their water solubility, ensuring greater stability and quality of the end products. This review presents a succinct overview of the available scientific evidence of the biological activity and toxicity of Pterodon species in addition to other important aspects, including phytochemical and technological features.

RESUMO

An oleaginous fraction obtained from an alcohol extract of the fruit of Pterodon pubescens Benth. (FHPp) was microencapsulated in polymeric systems. These systems were developed using a complex coacervation method and consisted of alginate/medium-molecular-weight chitosan (F1-MC), alginate/chitosan with greater than 75% deacetylation (F2-MC), and alginate/low-molecular-weight chitosan (F3-MC). These developed systems have the potential to both mask the taste of the extract, and to protect its constituents against possible chemical degradation. The influence of the formulation parameters and process were determined by chemical profiling and measurement of the microencapsulation efficiency of the oleaginous fraction, and by assessment of microcapsule morphology. The obtained formulations were slightly yellow, odorless, and had a pleasant taste. The average diameters of the microcapsules were 0.4679 µm (F2-MC), 0.5885 µm (F3-MC), and 0.9033 µm (F1-MC). The best formulation was F3-MC, with FHPp microencapsulation efficiency of 61.01 ± 2.00% and an in vitro release profile of 75.88 ± 0.45%; the content of vouacapans 3-4 was 99.49 ± 2.80%. The best model to describe the release kinetics for F1-MC and F3-MC was that proposed by Higuchi; however, F2-MC release displayed first-order kinetics; the release mechanism was of the supercase II type for all formulations.

Fruits of Pterodon pubescens Benth have been used traditionally for the treatment of rheumatism, sore throat, and respiratory disorders, and also as anti-inflammatory, analgesic, depurative, tonic, and hypoglycemic agent. The study was aimed at evaluating the anti-inflammatory activity of the hexane fraction of an ethanolic extract of P. pubescens fruits. The oil from P. pubescens fruits was extracted with ethanol and partitioned with hexane. The anti-inflammatory activity was measured with increasing doses of the hexane fraction (FHPp) by using a carrageenan-induced rat model of pleurisy and a rat model of complete Freund's adjuvant-induced arthritis by using an FHPp dose of 250 mg/kg for 21 days. Treatment with an FHPp resulted in anti-inflammatory activity in both models. The results of biochemical, hematological, and histological analyses indicated a significant decrease in glucose, cholesterol, and triglycerides levels (18.32%, 34.20%, and 41.70%, resp.) and reduction in the numbers of total leukocytes and mononuclear cells. The FHPp dose of 1000 mg/kg induced no changes in behavioral parameters, and no animal died. The results of this study extend the findings of previous reports that have shown that administration of extracts and fractions obtained from species of the genus Pterodon exhibits anti-inflammatory activity and lacks toxicity.

RESUMO

The crude hydroalcoholic extract from fruits of P. pubescens is widely used because of its anti-rheumatic, antinociceptive and anti-inflammatory activities. Furanoditerpenes have a vouacapan skeleton and are involved with the pharmacological activity of the oil extracted from P. pubescens fruits. Furanoditerpenes methyl 6α-acetoxy-7ß-hydroxyvouacapan-17ß-oate and methyl 6α-hydroxy-7ß-acetoxyvouacapan-17ß-oate from P. pubescens were isolated and identified. The present study developed and validated a GC-MS-SIM method for the separation and quantification of vouacapan constituents in a semipurified extract from P. pubescens fruits. The GC-MS analyses were carried out using a system equipped with a HP-5 capillary column (30 m × 0.25 mm). Temperature program: 100°C (4°C min(-1))-270°C (5 min), injector 260°C, detector 270°C. Helium was used as the carrier gas (0.7 bar, 1 mL min(-1)). The MS was taken at 70 eV. Scanning speed was 0.5 scans s(-1), from 50 to 650. Sample volume was 1 µL. Split 1:20. Analyses for validation of methodology were conducted by GC-MS-SIM (Single Ion Monitoring), where the ions monitored were 131, 145 and 146 (between 43 and 44.5 min), 105, 145 and 197 (from 44.5 to 45.3 min) and 131, 178 and 312 (from 45.3 to 48.5 min).Validation of the analytical method was based on the following parameters: linearity, robustness, limits of detection and quantification, precision (within-day and between-day variabilities), recovery and stability. The method was linear over a range of 12.81-2.56 µÎ³µΛ(-1) of vouacapan 1, 112.78-22.56 µg mL(-1) of vouacapan 2, and 333.34-66.67 µg mL(-1) of vouacapans 3 and 4, with detection limits of 0.39, 3.45 and 9,44 µg mL(-1) and quantification limits of 1.19, 10.47 and 28.62 µg mL(-1), respectively. Recovery values were 100.69%, 97.48% and 96.98% for vouacapans 1, 2 and 3-4, respectively. Thus, the method was efficient to separate and quantify furanoditerpenes in the extract or fraction.

RESUMO

In the course of an infection, the formation of reactive oxygen species by phagocytes and the antioxidant defense mechanisms of microorganisms play a crucial role in pathogenesis. In this study, isolates representing 8 pathogenic Candida species-Candida albicans, Candida dubliniensis, Candida famata, Candida glabrata, Candida guilliermondii, Candida krusei, Candida parapsilosis and Candida tropicalis-were compared with regard to their resistance to oxidative stress in vitro. We evaluated degree of resistance, induction of oxidative damage, capacity to adapt, and induction of antioxidant enzymes. The species showed variable sensitivity to oxidative attack. C. albicans, C. glabrata, and C. krusei were more resistant to oxidative stress under the conditions tested; C. parapsilosis and C. tropicalis presented medium resistance; and C. dubliniensis, C. famata, and C. guilliermondii were more sensitive. The overall greater resistance to oxidative stress of C. albicans and C. glabrata may provide an advantage to these species, which are the major causative agents of candidiasis.
